Caffeine, potassium sorbate, and sodium benzoate are common additives used in the production of soft drinks, energy drinks, and various food products. Accurate quantification of these compounds is crucial for quality control and regulatory compliance in the beverage industry. High-Performance Liquid Chromatography (HPLC) provides a robust method for the analysis of caffeine, potassium sorbate, and sodium benzoate mixtures. In this study, we describe the HPLC analysis of this mixture utilizing an Amaze HA mixed-mode column. Caffeine is a naturally occurring alkaloid found in coffee beans, tea leaves, and cacao pods. It is a white, crystalline powder with a bitter taste and acts as a stimulant, enhancing alertness and reducing fatigue. Potassium sorbate is a potassium salt of sorbic acid, a compound naturally found in some berries. It is widely used as a preservative in food and beverage products due to its ability to inhibit the growth of molds, yeasts, and bacteria. Sodium benzoate is the sodium salt of benzoic acid, a common food preservative found naturally in fruits. It is utilized in the food and beverage industry to prevent microbial growth and extend shelf life. The Amaze HA mixed-mode column features a unique stationary phase that combines hydrophobic, anion-exchange and cation-exclusion properties. The stationary phase consists of hydrophobic alkyl chains with embedded hydroxyl and amine groups, providing both reverse-phase and ion interactions. This mixed-mode chromatography enables enhanced selectivity and resolution of analytes, making it suitable for the analysis of polar and non-polar compounds, such as caffeine, potassium sorbate, and sodium benzoate. The Amaze HA column offers exceptional versatility and robustness, making it well-suited for various applications in the food and beverage industry. This method outlines the High-Performance Liquid Chromatography (HPLC) analysis of phytic acid using an Amaze HD HILIC mixed-mode column. The separation is performed in both Hydrophilic Interaction Liquid Chromatography (HILIC) and anion-exclusion modes to ensure comprehensive characterization of phytic acid. Amaze HD column can also be used in HILIC and cation-exchange mode for analysis of polar basic and polar neutral compounds. Anion-exclusion mode for phytic acid is chosen to achieve easy elution and perfect peak shape compared to analysis of this compound in HILIC and anion-exchange modes. Phytic acid, also known as inositol hexakisphosphate (IP6), is a naturally occurring compound found abundantly in plant seeds. It is a phosphorylated derivative of inositol and serves various roles, including as a storage form of phosphorus in plants and as an antioxidant in human diets. Phytic acid is highly water-soluble and negatively charged due to its multiple phosphate groups. Its complex chemical structure presents challenges for chromatographic analysis, necessitating specialized separation techniques like HILIC and anion-exclusion chromatography. The Amaze HD HILIC mixed-mode column is chosen for its versatility in handling polar and charged analytes like phytic acid. In HILIC mode, the hydrophilic component of the column facilitates retention and separation of polar compounds based on their hydrophilicity, while in anion-exclusion mode, the column selectively excludes charged species, allowing for separation based on differences in charge and size. By employing this HPLC method, accurate quantification and characterization of phytic acid can be achieved, facilitating its analysis in various food, pharmaceutical, and environmental samples.

Sucrose, fosfomycin, sodium and TRIS are part of the fosfomycin formulation. Sucrose, fosfomycin, sodium and tris are very polar in nature. Fosfomycin is a broad-spectrum antibiotic which contains an epoxy fragment as well as phosphonic acid. This makes this drug very polar and acidic. Sodium ion and tromethamine are used as counter-ions for this polar drug molecule. All four compounds are different in nature - sucrose is polar and neutral, sodium is polar and basic, TRIS is polar and basic, and fosfomycin is polar and acidic. It is hard to retain and separate these compounds in RP chromatography. HILIC can retain these polar compounds and mixed-mode HILIC on the Amaze TH column allows to introduce cation- and anion-exchange mechanisms for better retention and better retention control. The presence of ionic interaction, in addition to HILIC, allows better control of elution and retention. The retention time can be controlled by three parameters - amount of acetonitrile, amount of buffer and the buffer pH. This method can be used for the analysis of drug formulation and level of fosfomycin in biofluids (serum, blood, urine, etc). The method is robust, reproducible and compatible with all major detection techniques.

Ability to retain and separate compounds of opposite charge on one column with one method is important for analysis of salts, drugs, waste waters and other samples. We have developed a method for analysis of sodium and sulfate ions on Amaze TH column. Amaze TH column is HILIC mixed-mode column with both positive and negative sites on the surface. Addition of ionizable groups on the surface allows to increase a polarity of the stationary phase and also make these sites available for ion-exchange interactions. Both ions are well retained with good peak shape. Column and method can be used for analysis of other cations and anions in one run. Method is LC/MS compatible and provides unique selectivity and separation ability for polar, acidic and basic organic and inorganic compounds
